大鼠血浆中胆汁酸、胆红素及牛磺酸的LC-MS/MS分析

Determination of bile acids, bilirubin and taurine in rat plasma by LC-MS/MS

  • 摘要: 建立大鼠血浆中胆汁酸、胆红素和牛磺酸的LC-MS/MS同时测定方法。血浆样品经甲醇沉淀蛋白后LC-MS/MS分析,色谱柱为Zorbax SB-C18(150 mm×2.1 mm,5 μm),流动相为甲醇-10 mmol/L醋酸铵溶液(pH 3.0)梯度洗脱;检测系统采用电喷雾离子源(ESI)-三重四极杆质谱仪于负离子模式下进行选择性反应检测(SRM)。方法学验证结果表明,建立的方法灵敏度高、专属性好,可用于血浆样品中胆汁酸、胆红素和牛磺酸的含量测定。

     

    Abstract: A high-performance liquid chromatography tandem mass spectrometry(LC-MS/MS)method was developed for the simultaneous determination of bile acids, bilirubin and taurine in rat plasma. Plasma samples were protein precipitated with methanol and analyzed by LC-MS/MS. The analytes were well separated on a Zorbax SB-C18 column(150 mm×2. 1 mm, 5 μm)with gradient elution of methanol and 10 mmol/L ammonium acetate in aqueous solution(pH 3. 0). Detection was carried out on a triple quadrupole tandem mass spectrometer by selected reaction monitoring(SRM)in the negative ion mode. The proposed method proved to be sensitive and selective, which is suitable for the determination of bile acids, bilirubin and taurine in rat plasma.
