RP-HPLC法测定栀子大黄汤中多种成分的含量及其在方剂配伍研究中的应用
Quantitation of multiple constituents in Zhi-Zi-Da-Huang decoction by RP-HPLC and its application in the study of chemical components in herb compatibility

摘要: 在前期对经方栀子大黄汤中的化学成分进行定性研究基础上,本研究首次对栀子大黄汤中的9种特征成分进行定量测定,并将方法应用于分析该经方配伍前后成分的变化。试验采用HPLC-UV方法,同时测定栀子大黄汤水煎液中栀子苷、柚皮苷、橙皮苷及新橙皮苷的含量;并建立HPLC-FLD方法,针对该汤剂中芦荟大黄素、大黄酸、大黄素、大黄酚及大黄素甲醚的含量进行测定,使检测更为灵敏准确。最终所采用的方法经考察可以准确测定栀子大黄汤中上述9种特征成分的含量。对汤剂的测定结果显示,不同配伍组中各成分的含量发生了不同程度的变化,其中栀子大黄汤全方配伍能够增加栀子苷、柚皮苷、橙皮苷及新橙皮苷的含量,枳实-大黄配伍可以增加芦荟大黄素、大黄酸、大黄素和大黄素甲醚的含量,各配伍组大黄酸的含量均比单味大黄有明显增加。Abstract: Nine representative compounds in Zhi-Zi-Da-Huang decoction(ZZDHD)was quantitatively determined for the first time and their changes in herb compatibility was investigated on the basis of the authors′ previous qualitative researches of ZZDHD. Geniposide, naringin, hesperidin and neohesperidin were simultaneously determined by an HPLC-UV method. While an HPLC-FLD method was applied for the determination of aloe emodin, rhein, emodin, chrysophanol and physcion, in order to obtain better sensitivity and accuracy. The final methods were validated for the accurate determination of the 9 components. The results showed that the dissolution of geniposide, naringin, hesperidin and neohesperidin was promoted by the compatibility of ZZDHD, that the dissolution of anthraquinones was increased by the compatibility of Aurantii Fructus and Rhei Radix et Rhizoma, and that the contents of rhein all increased in ZZDHD and related drug pairs.
