Simultaneous determination of seven C21steroidal saponins in the radix of Cynanchum otophyllum Schneid.by HPLC wavelength switching method

  • 摘要: 建立HPLC波长切换法,同时测定青阳参中7个C21甾体皂苷的含量。采用Alltima C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-水为流动相,梯度洗脱,流速为1.0 mL/min,柱温为35 ℃,检测波长为259 nm(0~27 min),223 nm(27~72 min)。结果表明,该7个C21甾体皂苷在各自检测范围内线性良好,加样回收率范围在98.5%~103.9%之间。通过对不同产地的7个批次青阳参中7个成分的含量测定,认为该方法简便、准确、快速,可用于青阳参药材的质量控制。
    Abstract: An HPLC method was established to evaluate the quality of Cynanchum otophyllum with a simultaneous determination of seven C21 steroidal saponins. The method was carried out on an Alltima C18 column(250 mm×4. 6 mm, 5 μm)with a mobile phase consisting of acetonitrile-water at the flow rate of 1. 0 mL/min. The detection wavelength was 259 nm(0-27 min), 223 nm(27-72 min)and column temperature was 35 ℃. The seven compounds were completely separated with good linearity. The average recoveries varied from 98. 5% to 103. 9%. These results indicated that the method was convenient, accurate and rapid for the quality control of Radix of Cynanchum otophyllum in 7 commercial samples.
