Effects of estradiol valerate on the estrogen receptor expression in uterus, vagina and mammary gland of ovariectomized rats

  • 摘要: 以去卵巢大鼠为研究模型,观察戊酸雌二醇对去卵巢大鼠子宫、阴道、乳腺组织病理变化及雌激素受体α(ERα)和雌激素受体β(ERβ)表达的影响,探讨其雌激素样作用机制。采用HE染色法观察去卵巢大鼠组织的病理改变;采用免疫组织化学和Western blot技术检测子宫、阴道、乳腺ERα和ERβ蛋白的表达水平。结果显示,戊酸雌二醇可明显拮抗去卵巢大鼠靶器官的萎缩,其机制可能与上调靶器官雌激素受体(ERs)表达有关。
    Abstract: The objective of this study was to evaluate the effects of estradiol valerate(EV)on the estrogen receptor expression and the pathological changes of tissues in uterus, vagina and breast of ovariectomized rats, and investigate the estrogen-like mechanism. HE method was used to determine the tissue pathological changes; the protein expression levels of estrogen receptor alpha(ERα)and estrogen receptor beta(ERβ)in mammary gland, uterus and vagina were detected by immunohistochemistry and Western blot. The results indicated that estradiol valerate could obviously antagonize the atrophy of the target organs in ovariectomized rats, and the up-regulated expression of the estrogen receptors in target organs is the possible mechanism of EV induced estrogenic activity.
