Synthesis of(Z)-methyl-2-acetamidocinnamate

  • 摘要: (Z)-2-乙酰氨基肉桂酸甲酯是不对称催化氢化中用于评价手性催化剂的标准底物之一,但现有的合成方法都需要柱色谱纯化,而且收率低。本研究对其合成工艺进行改进,最终一步以NN′-二环己基碳二亚胺(DCC)为脱水剂和4-二甲胺基吡啶(DMAP)为催化剂,经简单地洗涤、萃取等操作,即可快速高效地得到高纯度的(Z)-2-乙酰氨基肉桂酸甲酯,产率可达68.5%。
    Abstract: In the investigation of asymmetric catalytic hydrogenation reaction, (Z)-methyl-2-acetamidocinnamate is often used as a standard substrate to assess the chiral catalysts. However, its previous syntheses suffered from low yield, relying on column chromatography for isolation. Herein, (Z)-methyl-2-acetamidocinnamate was obtained with 68. 5% isolation yield via simple operations such as wash and extraction using dicyclohexylcarbodiimide(DCC)as dehydrant and 4-dimethylaminopyridine(DMAP)as catalyst in the last synthetic step.
