虫草多糖CPS-A对血管紧张素Ⅱ诱导肝L02细胞损伤的保护作用

Protective effects of Cordyceps sinensis polysaccharide CPS-A on angiotensin II-induced injury of liver L02 cells

  • 摘要: 探讨虫草多糖CPS-A对Ang Ⅱ诱导的人肝L02细胞损伤的保护作用。MTT法考察Ang Ⅱ、CPS-A对L02细胞增殖的影响,Ang Ⅱ刺激L02细胞后,考察CPS-A对L02细胞的损伤保护作用;PCR、Real-Time PCR和Western blot法检测IL-6、IL-1β、AT1R、AT2R、NF-κB p65、TNFα等因子mRNA和蛋白质的表达水平。结果表明:Ang Ⅱ、CPS-A有效抑制L02细胞增殖的浓度分别为1×10-5 mol/L和200 μg/mL,PCR、Real-Time PCR和Western blot结果显示,CPS-A可以显著下调IL-6、IL-1β、TNF-α、NF-κB和AT1R的表达。CPS-A对Ang Ⅱ诱导的L02细胞损伤具有很好的保护作用。

     

    Abstract: This study aimed at the molecular mechanism of Cordyceps sinensis polysaccharide-A(CPS-A)on angiotensin(Ang II)-induced injury of L02 cells. The effect of AngII and CPS-A on the proliferation of L02 cells was analyzed by MTT assay. PCR, Real-Time PCR and Western blot were also employed to determine the expression of IL- 1β, AT1R, AT2R, NF-κB p65, TNFα and other inflammatory factors at mRNA and protein levels. The results showed that AngⅡ and CPS-A could inhibit the proliferation of L02 cells by 1×10-5 mol/L and 200 μg/mL, respectively. PCR, Real-Time PCR and Western blot showed that CPS-A could significantly down-regulate IL-1β, TNF-α, NF-κB and AT1R. CPS-A has a good protective effect on AngII-induced L02 cell injury.
