胰脂肪酶抑制剂筛选方法的研究进展

Research advances in screening methods for pancreatic lipase inhibitors

  • 摘要: 肥胖症与高血脂症、动脉粥样硬化、2型糖尿病等多种代谢性疾病密切相关。胰脂肪酶是将食源性脂质转化成甘油三酯和脂肪酸的关键酶;其活性的有效抑制已成为治疗肥胖症的重要策略。本文系统阐述了游离胰脂肪酶筛选法、固定化胰脂肪酶筛选法和虚拟筛选法的筛选原理、优缺点、研究现状及影响因素,并对“虚拟筛选-固定化酶富集-游离酶验证”的多维集成筛选范式进行展望,以期为高效低毒的胰脂肪酶抑制剂开发提供参考。

     

    Abstract: Obesity is closely associated with a variety of metabolic diseases, including hyperlipidemia, atherosclerosis, and type 2 diabetes mellitus. Pancreatic lipase plays a critical role in the conversion of dietary lipids into triglycerides and free fatty acids, and the effective inhibition of its activity has emerged as a key therapeutic strategy for obesity management. This article systematically elucidates the principles, advantages, limitations, research progress, and influencing factors of several screening methods for pancreatic lipase inhibitors, including free enzyme-based assays, immobilized enzyme-based assays, and virtual screening. Furthermore, it offers a perspective on a multi-dimensional integrated screening paradigm characterized by "virtual screening-enrichment via immobilized enzymes-validation with free enzymes, " with the aim of providing valuable insights for the development of efficient and low-toxicity pancreatic lipase inhibitors.
