溴化乙锭对吸水链霉菌产生除莠霉素的影响

Effect of Ethidium Bromide on Production of Herbimycin A in Streptomyces hygroscopicus NAP-05

  • 摘要: 从南京土壤中分离到一株吸水链霉菌,命名为NAP-05,该菌株能产生除莠霉素,并且在菌体细胞内存在另一种初步鉴定为聚醚类抗生素的物质。该菌还能分泌淀粉酶。用嵌入染料如溴化乙锭(EB)等处理该菌株,发现EB处理株大部分仍保持产抗能力,而用吖啶橙(AO)处理亲株后,大部分的处理株失去除莠霉素产生能力。用原生质体再生方法处理NAP-05,发现多数再生株的产抗能力下降,提示NAP-05菌株的除莠霉素生物合成调节可能受到染色体外因子的影响。

     

    Abstract: Ethidium bromide(EB) and acridine orange were used to treat on the Streptomyces hygroscopicus NAP-05, which produces herbimycin and amylase. After treatment, the most mutants lose the herbimycin production, while most EB mutants still produce the antibiotics. Protoplost regeneration was carried out in the organism NAP-05. The most regeneration strains lose or decrease the herbimycin producing ability.
