手性高效液相色谱法拆分硝酸芬替康唑对映异构体

Separation of fenticonazole nitrate enantiomers by chiral-HPLC

  • 摘要: 建立硝酸芬替康唑对映异构体的手性色谱分析方法。通过考察手性固定相、流动相比例以及柱温等因素对硝酸芬替康唑对映异构体分离度的影响,进行了色谱分离条件的优化,最终确定的色谱条件为:Daicel AY-H手性色谱柱(250 mm×4.6 mm,5 μm),正己烷-异丙醇-二乙胺(85∶15∶0.15)为流动相,检测波长为254 nm,流速为1.0 mL/min,柱温35 ℃。结果在该色谱条件下,(-)-硝酸芬替康唑与其对映异构体分离良好,分离度大于1.5。建立的方法专属性强,操作快速、简单,适用于硝酸芬替康唑对映异构体的测定和质量评价。

     

    Abstract: A chiral-HPLC method was established to separate and determine the fenticonazole nitrate enantiomers.The factors such as chiral stationary phases,different proportion of mobile phase and column temperatures were investigated.Finfally,a Daicel AY-H analytical column (250 mm×4.6 mm,5 μm) was selected with the n-hexane:isopropanol:diethylamine (85∶15∶0.15) as the mobile phase at a detection wavelength of 254 nm and a flow rate of 1.0 mL/min.The column temperature was 35 °C.Under this chromatographic conditions,(-)-fenticonazole nitrate could be completely separated with its enantiomer and the resolution was not less than 1.5.The method was proved to be simple,sensitive and enantioselective,and it is suitable for determination and evaluation of fenticonazole nitrate enantiomers.
