新型抗HIV药物靶点核糖核酸酶H抑制剂的研究进展

摘要: 随着艾滋病感染人数和HIV耐药性突变的不断增加,对抗HIV新药需求越来越强烈。目前抗HIV新药研究主要分两方面进行,一方面是针对现有靶点开发新药,另一方面致力于寻找新的药物靶点。核糖核酸酶H是逆转录酶上具有自身独特酶功能的催化区域,成为抗HIV药物研究的一个新靶点。本文主要对近年来发现的核糖核酸酶H抑制剂进行总结,阐述其作用机制,为进一步深入开展核糖核酸酶H抑制剂的改造和设计提供一些信息。Abstract: As the infection of the acquired immunodeficiency syndrome (AIDS) and the mutation of the HIV virus keep increasing,there’s an urgent demand for developing new anti-HIV drugs.Up till now,the research on anti-HIV agents has been carried out in two ways:based on known targets or new targets.Ribonuclease H (RNase H),which has with its own unique catalytic sites of enzyme function and is regarded as a novel drug target for anti-HIV therapy,is a domain of the reverse transcriptase.This review summarizes RNase H inhibitors discovered in recent years and explains their mechanisms of action which aims at providing some useful information for further structural modifications and design of RNase H inhibitors.
