蛇毒三肽pENW对FeCl3诱导的大鼠动脉血栓形成的抑制作用及其机制

Inhibition of pENW on arterial thrombosis induced by FeCl3 in rat

  • 摘要: 采用FeCl3诱导的大鼠动脉血栓模型考察蛇毒三肽焦谷氨酸-天冬酰胺-色氨酸(pENW)的抗血栓形成作用,并对其抗血栓作用进行了初步的机制探讨。应用Born比浊法和Grynkiewicz方法分别测定pENW对大鼠血小板聚集功能和血小板内钙水平的影响,通过凝血三项(活化部分凝血活酶时间、凝血酶原时间、凝血酶时间)考察pENW对凝血系统的影响;实验结果表明,静脉注射pENW(iv)能抑制FeCl3诱导的大鼠动脉血栓形成(P<0.05),抑制ADP诱导的血小板聚集作用(P<0.05),抑制ADP诱导的血小板胞浆内游离Ca2+浓度([Ca2+i)升高,但对凝血酶系统没有显著性影响;实验结果提示,pENW具有抗血栓形成作用,其机制可能与抑制血小板聚集、抑制血小板胞浆[Ca2+i升高有关。

     

    Abstract: The model of arterial thrombosis induced by FeCl3 in rat was used to investigate the antithrombotic effect of pGlu-Asn-Trp(pENW).Platelet aggregation was tested according to Born’s methods.Platelet cytosolic calcium was determined using of Fura-2/AM.Prothrombin time (PT),thrombin time (TT) and activated partial thromboplastin time (APTT) were tested for exploring the effect of pENW on prothrombin.pENW (iv) inhibited arterial thrombosis induced by FeCl3 and platelet aggregation induced by ADP,respectively (P<0.05).Ex vivo pENW markedly lowered the cytosolic calcium higher concentration induced by ADP in platelet.However,there was no significant effect on PT,TT and APTT.These results showed that pENW evidently inhibited platelet aggregation and decreased the concentration of platelet cytosolic calcium.Results suggest that the antithrombotic effect of pENW may be related to its anti-platelet aggregation and the decrease of platelet cytosolic calcium.
