竞争性内源RNA(ceRNAs)研究进展——RNA对话的新机制

Progress in research of competing endogenous RNA:a novel RNA regulation mechanism

  • 摘要: microRNAs(miRNAs)是一类长度约为22 nt的非编码单链小RNA分子,通过其种子序列与靶基因的3′UTR互补并引导沉默复合体(RISC)降解或抑制靶基因的翻译,在转录后水平调控靶基因的表达。一个miRNA可调控多个靶基因,同一个靶基因也可以受多个miRNA调控。最新的研究发现mRNA之间可以通过miRNAs反应原件(MREs)达到相互调控的目的,这种新的调控机制被称作竞争性内源RNA(ceRNAs)假说,这种RNA之间新的对话机制的出现不仅在转录组学水平赋予了mRNAs、长链非编码RNA及假基因的转录产物新的生物学功能,扩大人类基因组中的功能性遗传信息,并且绘制了一个更庞大的miRNAs与mRNA相互调控的网络,为深入研究肿瘤等疾病的发病机制提供新的理论依据。本文从ceRNAs调控网络的成员、ceRNAs调控机制的作用基础、作用方式及ceRNAs与肿瘤等多方面对ceRNAs调控机制的研究进展进行综述。

     

    Abstract: MicroRNAs (miRNAs),a series of endogenous,non coding and single strand RNA approximately containing 22 nucleotides,regulate messenger RNAs at the post transcriptional level through binding to complementary sequences in un translated regions (UTR),inducing RNA induced silencing complex (RISC) to degrade mRNA or repress translationA miRNA can have multiple mRNA targets,and the same mRNA can be regulated by many miRNAsLatest researches show that RNAs regulate each other using microRNA response elements (MREs) and this mechanism is known as "competing endogenous RNA" (ceRNA)ceRNAs hypothesis has enabled new features for mRNA and activity forms a large scale regulatory network across the transcriptome,greatly expanding the functional genetic information in the human genome and playing important roles in pathological conditions,such as cancerThis paper reviewed ceRNAs regulating mechanism with the following respects:members of ceRNAs regulatory network,ceRNAs regulating bases,the mechanismm of ceRNAs as well as ceRNAs with tumor.
