降压益肾颗粒对自发性高血压大鼠肾组织细胞外基质降解系统的影响

Effect of the Jiangya Yishen granule on regulative system of extracellular matrix decomposition of spontaneously hypertensive rats

  • 摘要: 采用实时荧光PCR和激光共聚焦技术研究降压益肾颗粒(JYYS)对自发性高血压大鼠(SHR)肾组织细胞外基质降解系统转化生长因子-β1(TGF-β1)、Ⅳ型胶原蛋白(COLⅣ)、基质金蛋白酶-9(MMP-9)及其抑制物金属蛋白酶组织抑制因子-1(TIMP-1)表达的影响。以JYYS和苯那普利为研究对象,考察不同剂量JYYS和苯那普利对SHR肾脏TGF-β1、COLⅣ、MMP-9及其抑制物TIMP-1 mRNA表达和蛋白表达的差异。结果表明:JYYS长期给药可降低TIMP-1、TGF-β1及COLⅣ转录和蛋白水平,上调肾组织局部MMP-9mRNA和蛋白水平,从而纠正MMP-9/TIMP-1失衡,促进细胞外基质的降解,减少细胞外基质的积聚,产生肾脏保护作用。

     

    Abstract: This article investigate the effect of the Jiangya Yishen(JYYS) granule on regulative system of extracellular matrix(ECM)decomposition of spontaneously hypertensive rats(SHR),we determined the expression of include transforming growth factor β1(TGF-β1),collagen type IV(COLIV),matrix metalloproteinase-9(MMP-9),tissue inhibitor of metalloproteinase-1(TIMP-1)by utilizing real time fluorescent PCR technique (RT-PCR) and laser confocal fluorescence microscopy.The results showed that JYYS could enhance the level of renal MMP-9 mRNA and its protein level,meanwhile down regulate the transcription of TIMP-1,TGF-β1,COLIV and decreasing their protein level after long term treatment.Therefore,JYYS corrected the imbalance of MMP-9/TIMP-1,then enhanced the degradation of ECM and decreased the accumulation of ECM to protect kidney in SHR.
