Quantitation of multiple constituents in Zhi-Zi-Da-Huang decoction by RP-HPLC and its application in the study of chemical components in herb compatibility

Abstract
Nine representative compounds in Zhi-Zi-Da-Huang decoction(ZZDHD)was quantitatively determined for the first time and their changes in herb compatibility was investigated on the basis of the authors′ previous qualitative researches of ZZDHD. Geniposide, naringin, hesperidin and neohesperidin were simultaneously determined by an HPLC-UV method. While an HPLC-FLD method was applied for the determination of aloe emodin, rhein, emodin, chrysophanol and physcion, in order to obtain better sensitivity and accuracy. The final methods were validated for the accurate determination of the 9 components. The results showed that the dissolution of geniposide, naringin, hesperidin and neohesperidin was promoted by the compatibility of ZZDHD, that the dissolution of anthraquinones was increased by the compatibility of Aurantii Fructus and Rhei Radix et Rhizoma, and that the contents of rhein all increased in ZZDHD and related drug pairs.
