Effect of COX/5-LOX dual inhibitor ZLJ-6 on the expression of LPS-induced inflammation factors in macrophage cells

Abstract
The anti-inflammatory effects of COX/5-LOX dual inhibitor ZLJ-6 in LPS-stimulated RAW 264.7 cells were investigated.RAW 264.7 cells were stimulated with lipopolysaccharide (LPS) 1 μg/mL to set up the cell inflammatory model;various concentrations (3,10 and 30 μmol/L) of ZLJ-6 were administered to explore its effect on the production of NO,TNF-α and IL-6,and the expression of COX-2 and inducible nitric oxide synthase (iNOS) in these LPS-stimulated cells.The production of inflammatory factor nitric oxide (NO),tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in the supernatant was measured by Griess reagent and ELISA respectively.Western blotting was also applied to detect the expression of intracellular COX-2 and iNOS.The results showed that levels of NO,IL-6 and TNF-α in the supernatant and expression of COX-2 and iNOS were dramatically higher after LPS stimulated RAW 264.7 cells.ZLJ-6 decreased the production of NO,TNF-α and IL-6 in a concentration-dependent manner.The expression of COX-2 and iNOS in LPS-stimulated RAW264.7 cells were also decreased by treatment with ZLJ-6.These results indicated that ZLJ-6 can inhibit the inflammation response in LPS-stimulated RAW264.7 cell,which might be mediated by decreasing the production of NO,TNF-α and IL-6 and down-regulating the expression of COX-2 and iNOS.
