Effects of active peptide extracted from cheese whey on the expression of PPARγ mRNA and NF-κB activity in 3T3-L1 adipocytes

  • This study was to explore the effect of active peptides extracted and isolated from traditional Xinjiang fermented cheese whey(whey active peptide)on the expression of PPARγ mRNA and activities of NF-κB in the inflammatory state of 3T3-L1 preadipocyte.3T3-L1 were cultured and induced into maturation,then stimulated with LPS to induce the inflammatory state,given at different concentrations of whey active peptides (1,10,20,50 mg/L),and positive control group HDL(10,50,100 mg/L),then the expression of PPARγ mRNA,and the activity of NF-κB were measured after the cells were collected.PPARγ mRNA were highly expressed in the matured 3T3-1 adipocyte stimulated with LPS.Compared with LPS group,the expression of PPARγ mRNA were increased significantly in 50,100 mg/L HDL group and 20,50 mg/L active peptide group.The activity of NF-κB were increased significantly in LPS group compared with the control group,while decreased significantly in 50,100 mg/L HDL group and 20,50 mg/L active group compared with LPS group.Whey active peptides can up-regulate the expression of PPARγ mRNA and decrease the NF-κB activity.
