Effects of active peptide extracted from cheese whey on the expression of PPARγ mRNA and NF-κB activity in 3T3-L1 adipocytes

Abstract
This study was to explore the effect of active peptides extracted and isolated from traditional Xinjiang fermented cheese whey(whey active peptide)on the expression of PPARγ mRNA and activities of NF-κB in the inflammatory state of 3T3-L1 preadipocyte.3T3-L1 were cultured and induced into maturation,then stimulated with LPS to induce the inflammatory state,given at different concentrations of whey active peptides (1,10,20,50 mg/L),and positive control group HDL(10,50,100 mg/L),then the expression of PPARγ mRNA,and the activity of NF-κB were measured after the cells were collected.PPARγ mRNA were highly expressed in the matured 3T3-1 adipocyte stimulated with LPS.Compared with LPS group,the expression of PPARγ mRNA were increased significantly in 50,100 mg/L HDL group and 20,50 mg/L active peptide group.The activity of NF-κB were increased significantly in LPS group compared with the control group,while decreased significantly in 50,100 mg/L HDL group and 20,50 mg/L active group compared with LPS group.Whey active peptides can up-regulate the expression of PPARγ mRNA and decrease the NF-κB activity.
