Effect of the ethyl acetate extract of Farfarae Flos on inflammatory factors

  • The effect and mechanism of the ethyl acetate extract of Farfarae Flos (FF-EtOAc) on inflammatory factors in LPS-stimulated RAW 264.7 model was studied in this paper.The levels of NO,TNF-α,and IL-6 were tested by Griess assay and ELISA,and NF-κB binding protein was tested by Western blot.Results showed that higher levels of NO,IL-6 and TNF-α were released,and the level of phosphorylation of NF-κB P-65 was higher than normal group;both FF-EtOAc and sodium salicylate had inhibitory effect on NO,IL-6 and TNF-α production in LPS-stimulated RAW 264.7 cells.Western blot results demonstrated that FF-EtOAc can significantly inhibit LPS- induced phosphorylation of NF-κB P-65 in RAW 264.7.We inferred that the mechanisms of FF-EtOAc anti- inflammatory effect in LPS-stimulated RAW 264.7 may be through inhibiting of NF-κB P-65 phosphorylation to reduce the production of NO,TNF-α and IL-6.
