UPLC法同时测定胆木中6种成分的含量

Simultaneous determination of six main compounds in Nauclea officinalis by ultra-performance liquid chromatography

  • 摘要: 建立胆木药材中原儿茶酸、绿原酸、短小舌根草苷、3-表短小舌根、异常春花苷内酰胺和喜果苷同时测定的UPLC方法。采用Acquity BEH C18色谱柱(2.1 mm×100 mm,1.7 μm),乙腈-0.1%甲酸为流动相,梯度洗脱,流速为0.4 mL/min,进样量为2 μL,柱温为40 ℃的色谱条件。在选定的色谱条件下,6种成分均能达到良好分离,并对方法进行了系统的验证。结果表明,线性关系良好(R2>;0.999 6),日内、日间精密度RSD≤ 4.05%,加样回收率为96.59%~101.8%。本方法简便、快速、准确,可用于胆木药材的质量控制。

     

    Abstract: A UPLC method was established for the simultaneous determination of protocatechuic acid, chlorogenic acid, pumiloside, 3-epi-pumiloside, strictosamide and vincosamide in Nauclea officinalis. The experiment was performed with an Acquity BEH C18(2. 1 mm ×100 mm, 1. 7 μm)column and the mobile phase consisting of acetonitrile and 0. 1% formic acid with gradient elution. The flow rate of 0. 4 mL/min and the injection volume of 2 μL as well as the column temperature of 40 ℃ were employed. Under the above-mentioned condition, 6 components in Nauclea officinalis were separated completely and the method was fully validated. Good linearity(R2> 0. 999 6), precision of inter- and intra-day(RSD≤4. 05%)together with average recoveries of 96. 59%-101. 8% were obtained. The method was proved to be rapid and accurate and can be used for the quality control of Nauclea officinalis.
