人参茎愈伤组织的细胞悬浮培养

Suspension Culture of Stem Callus Cells of Panax Ginseng

  • 摘要: 以自旋式10升大瓶进行的人参细胞悬浮培养,一般生长速度为21—25克/升/日,如连续培养可提高到74克/升/日,最终得率为12克/升/日(千重)。改用简化培养液,使成本从500元/米~3降为100元/米~3,而且对细胞的产量和粗皂甙含量均无不良影响。在继代培养33个月后,生长量提高约10倍,而粗皂甙含量下降约3倍,但仍与人参药材含量相近。悬浮培养的液体中含有一定数量的粗皂甙。

     

    Abstract: Suspension culture of stem callus cells of Panax ginseng was carried out in 10L. rotating bottles. the growth rate of cells was 21-25g/L/day (fresh weight) and it could be increased to 74g/L/day by continuous culture.The final yield was 12g/L (dry weight). When a simplified culture medium was used, the production cost was reduced to 1/5, the yeild of cells and the crude ginsenoside content were not affected. In addition, it has been observed that when culture of successive generation continued for 33 months, the growth rate of cells was about 10 times heigher, but the crude ginsenoside content was decreased to about one-third, which is about the same as that of the crude drug (6.0%). The suspension culture medium also contained a certain amount of crude ginsenosides.
