薄层—紫外分光光度法测定尿中氢氯噻嗪含量

QUANTITATIVE DETERMINATION OF HYDROCHLOROTHIAZIDE IN URINE BY TLC-UV SPECTROPHOTOMETRY

  • 摘要: 薄层分离和紫外分光光度法测定尿中氢氯噻嗪含量的步骤是:用乙酸乙酯从尿中提取氢氯噻嗪并转溶于丙酮中,经用苯—乙醚—丙酮(3:3:4)展开剂进行硅胶G薄层层析,氢氧噻嗪在紫外分析仪下显荧光斑点,经无水乙醇洗脱,在紫外分光光度计λ272nm处测其含量。本法原点回收98.8%;展开回收97.4%;一例预试者口服氢氯噻嗪片48小时尿中累积回收72.30%。

     

    Abstract: The hydrochlorothiazide extracted from the urine by ethylacetate is dissolved inacetone. In the TLC, silica gel G is used as the adsorbent and the mixture ofbenzene-ether-acetone (3: 3: 4) is used as the developing solvent. The hydrochloro-thiazide spot, visualized under UV light in the chromatoplate, is eluted with absolute alcohol and determined at λ272nm by UV spectrophotometer. The recovery of hydrochlorothiazide from original spot and the spot after developing is 98.8% and 97.4% respectively. The total recovery of hydrochlorothiazide from the human urine accumulated in the period of 48 hrs after oral administration of this drug is 72.30%.
