应用改进的GLC法研究扑尔敏控释微丸的体内外相关性

AN IN VITRO AND IN VIVO CORRELATIVITY STUDY OF CHLORPHENIRAMINE CONTROLLEDRELEASE PELLETS BY MODIFIED GLC-ASSAY

  • 摘要: 本文改进了扑尔敏血药浓度的气相测定法。选用安定为内标,在异辛烷一次提取全血后用N~(63)电子捕获器检测。结果表明,在0.5~12ng/ml范围内,扑尔敏及安定的色谱峰面积之比(Ac/Ad)与血液中两者浓度之比(Cc Cd)有良好的线性关系(r=0.996),回收率在90%以上(平均偏差4%)。应用此法测定了人口服扑尔敏微丸及速释微丸的血药浓度,并由此计算得出控释微丸的体内吸收百分数(Pi)与体外溶出百分数(Po)间的相关方程式为Po=0.9597Pi 3.3225(r=0.916)
