葡萄球菌的原生质体融合及遗传转移

PROTOPLAST FUSION AND GENETIC TRANSFER OF STAPHYLOCOCCI

  • 摘要: 本文报道在金黄色葡萄球菌多剂抗生素耐药菌株(22)及白色葡萄球菌(Rif~r,Nal~r)间进行的原生质体融合试验。在高渗缓冲培养基中,将对数生长期中、后期的培养物,以溶菌酶及溶葡萄球菌素处理,DM_3培养基再生得原生质体。将二亲本葡萄球菌的原生质体混合,以CaCl_2及PEG(6000)处理使发生融合及种间遗传转移。根据重组子的色素、耐药性及在琼脂糖凝胶电泳中显示的质粒带进行遗传分析,证明在金黄色葡萄球菌及白色葡萄球菌中,质粒和染色体的遗传标记均可相互转移,再次证实在金黄色葡萄球菌(22)中存在分子量为2.0Mdal的pcr质粒。
