花粉多糖的分离纯化及其组成单糖分析

Isolation and Purification of Polysacchrides from Pollen of Rap and Analysis of Their Components

  • 摘要: 油菜花粉经破壁、热水提取,用三氯醋酸除杂蛋白、透析、乙醇沉淀获得粗品多糖。用CTAB络合法分离了花粉酸性多糖与中性多糖成分。中性多糖用Sephadex G-100凝胶柱层析进一步纯化得P-A、P-B、P-C 3个组分。酸性多糖用DEAE-Sephadex A-25纯化得P-D、P-E 2个组分,聚丙烯酰胺凝胶电泳均为单一区带。P-A、P-B、P-C、P-D、P-E的酸水解产物经气相色谱分析表明,均含L-岩藻糖、L-阿拉伯糖、D-木糖、D-甘露糖、D-半乳糖、D-葡萄糖及L-鼠李糖。而酸性多糖除含以上单糖组分外,还含有已糖醛酸,但不含硫酸基。
