毛细管电泳法分析血清中华法林对映体的浓度

Determination of Warfarin Enantiomers in Serum by Capillary Electrophoresis

  • 摘要: 建立了血中华法林对映体的毛细管电泳分析方法。电泳缓冲液由140mmol/L磷酸盐缓冲液和7mmol/L的甲基β-环糊精组成(pH7.9)。工作电压13kV,检测波长210nm。两种对映体S(-)-华法林及R(+)-华法林达到基线分离,在0~5μg/ml范围内呈线性。血中华法林对映体最低检测浓度均为0.02μg/ml,精密度及回收率均较好。

     

    Abstract: A method for the separation of warfarin enantiomers in serum by capillary electrophoresis was developed. The run buffer contained 140 mmol/L phosphate and 7 mmol /L methylated β-cyclo-dextrin(pH7.9), applied voltage 13kV, and detected at 210 nm. A baseline separation of two warfarin enantiomers can be obtained. The standard curve was linear from 0 to 5.0μg /ml and the limit ofdetection was 0.02μg/ml in serum. The precision and recovery rate was satisfying.
