大鼠胆汁中四种盐酸非洛普代谢物的HPLC测定法及其动态研究

Simultaneous Determination of Four Metabolites of Phenoprolamine Hydrochloride and Their Pharmacokinetics in Rat Bile by HPLC

  • 摘要: 建立了同时测定大鼠胆汁中 4种盐酸非洛普 ( DDPH) 相代谢物的 HPLC方法。胆汁样品经盐酸水解后以 Alltech C1 8固相萃取小柱纯化富集 ,然后进行 HPLC分析。色谱柱为 Hypersil ODS2 1 0μ 30 cm× 4 .6mm ID,流动相为甲醇 -水 -三乙胺 -冰乙酸 ( 450∶ 550∶ 1∶ 0 .7) ,检测波长为 2 78nm。测定了 ip DDPH后大鼠胆汁中 4个 DDPH代谢物的体内动态。结果表明 M1和 M2为 DDPH主要体内代谢产物。给药后 5h DDPH代谢产物的胆汁累积排泄量已达胆汁总排泄量的 75%~ 80 %。给药后 0 .5~ 1 h时间段 DDPH代谢物的胆汁排泄速率达峰值
