贯叶连翘提取物对单胺氧化酶与单胺递质的影响

Effect of Hypericum Extracts on Monoamine Oxidase and Monoamine Neurotransmitters

  • 摘要: 目的:探讨贯叶连翘提取物抗抑郁的作用机理。方法:测定药的对小鼠脑组织内的单胺氧化酶A和B(MAO-A和MAO-B)的活性,及用HPLC-电化学检测器测定对大鼠纹状体和前脑边缘区的单胺递质及其代谢产物含量。结果:贯叶连翘提取物(250mg/kg)对小鼠脑组织的MAO-B活性的抑制率为12.6%,50mg/kg对MAO-A和MAO-B活性的抑制率分别为13.2%和18.8%。250mg/kg组大鼠纹状体DOPAC的含量明显增加;500mg/kg组能增加大鼠边缘区的NA和DA,及代谢物DOPAC,HVA和T-HIAA的含量。同时,纹状体的DA与DOPAC和HVA均明显升高。结论:贯叶连翘提取物可能是通过抑制单胺氧化酶,以及抑制单胺类神经递质的摄取而发挥抗抑郁作用。
