HPLC-MSnanalysis of saponins in Rhizoma Anemarrhenae

  • 摘要: 目的 : 应用HPLC-MSn方法分析知母中的甾体皂苷成分。 方法 : 采用Hedera ODS-2色谱柱(250 mm×4.6 mm ID,5 μm),流动相:乙腈-水梯度洗脱,ELSD检测器,ESI离子源,正负离子模式下采集数据。 结果 : 根据负离子模式下的准离子峰和正离子模式下的二、三级质谱裂解规律,结合文献并与部分对照品比较,鉴定了15个甾体皂苷。 结论 : 该法选择性强、灵敏度高、操作简便,适用于知母甾体皂苷类化合物的定性分析。

     

    Abstract: Aim :To investigate the steroidal saponins of Rhizoma Anemarrhenae through HPLC-MSnanalysis. Methods :The chromatographic conditions were as follows:Hedera ODS-2(250 mm×4.6 mm ID,5 μm) column;water(A) and acetonitrile(B),gradient elute,ELSD detector,the ESI source was used and the data in negative and positive mode were acquired. Results :The molecular ions in negative ion modes and product ions in positive ion were observed for molecule mass information and steroidal skeleton,and the structures were elucidated in comparison with the reference data and the standards.Fifteen steroidal saponins were identified. Conclusion :The method is simple and rapid for the identification of the steroidal saponins from Anemarrhena asphodeloides Bge.
