  • 摘要: 为了探讨不同结构的美洛昔康微乳的透皮行为,以脂溶性药物美洛昔康为模型药物,以油酸-(氢化聚氧乙烯蓖麻油-乙醇)-水[表面活性剂与助表面活性剂的质量比(Km)=2]微乳相图中稀释线91上的一系列处方为研究对象,通过测定电导率、黏度和表面张力的变化确定微乳结构,分别测定不同结构微乳对药物的增溶能力,进而测定该系列微乳的经皮渗透系数,比较并分析微乳结构对其经皮渗透行为的影响。在联合运用多种方法确定了微乳的3种结构后,发现增溶能力以W/O型最强,其次为双连续型、O/W型;但在相同载药量的情况下,经皮渗透能力却以水包油型最强、其次为双连续相、油包水型。这一发现证明了微乳的结构对其载药能力和经皮渗透速度有较为显著的影响。

     

    Abstract: The purpose of this article was to investigate the effect of structure on the transdermal behavior of meloxicam microemulsion.Meloxicam microemulsions containing of oleic acid,Cremophor RH 40,ethanol and water(Km=2) were prepared,and the structures of the microemulsion formulations along dilution line 91 in the microemulsion phase diagram were determined by electrical conductivity,viscosity and surface tension.Moreover,the formulation improvement in the solubilization of meloxicam and the transdermal permeation coefficient of the microemulsions were evaluated.It was proved that W/O type microemulsion had the best solubilization potential,followed by B.C.and O/W,and that O/W microemulsion had the best permeation rate,followed by B.C and W/O type.Thus the structures of microemulsion had accounted for the solubilization potential and transdermal permeation.
