大承气汤水煎剂HPLC指纹图谱研究

  • 摘要: 利用中药指纹图谱研究大承气汤水煎剂,为该方的物质基础和质量控制提供了科学依据。采用Kromasil-C 18色谱柱,乙腈-0.05%磷酸水溶液为流动相,梯度洗脱,流速0.6 mL/min,检测波长261 nm,柱温30 ℃,以大黄酸为参照物,采用相关系数法对10批大承气汤水煎剂进行分析。建立了大承气汤水煎剂的HPLC指纹图谱,标定了19个共有峰,鉴定了其中7个峰,10批样品的相似度在0.977~0.997之间。研究表明:建立的方法准确、可靠,可用于大承气汤物质基础的研究和其中成药开发时的质量控制。

     

    Abstract: Fingerprinting provides a method to analyze the complex constituents in traditional Chinese prescriptions.Dachengqi decoction was,therefore,studied by HPLC fingerprint in order to offer the foundation for the study on the chemical material of this formula and the evidence for quality control of traditional Chinese drugs.Ten batches of Dachengqi decoction were measured by HPLC using Rhein as a reference substance and the chromatographic separation was performed on a Kromasil-C 18column. A linear gradient elution with A (acetonitrile) and B (0.05%H3PO4) was used. The detection wavelength was set at 261 nm; the flow rate was 0.6 mL/min; and the column temperature was 30 ℃.Nineteen common peaks were signed,seven identified. Their similarity threshold was 0.97. The established method is accurate and repeatable,and can be used for the study on the chemical material of this formula and the evidence for quality control of traditional Chinese drugs.
