用Fluo3-AM测定2型糖尿病患者淋巴细胞内Ca~(2 )浓度及药物对患者胞内Ca~(2 )浓度的影响

Determination of the Concentration of Intracellular Calcium in Lymphocyte by Fluo 3 AM and Study of the Influence of the Drugs on Calcium Release

  • 摘要: 用 Fluo 3- AM为荧光探针 ,测定 2型糖尿病患者淋巴细胞内 Ca2 浓度 ,观察了肾上腺素、多巴酚丁胺、Bay K 864 4、胰岛素等药物对胞内 Ca2 的作用机制。实验表明患者胞浆内 Ca2 浓度与正常人相比显著升高 ;药物对胞内 Ca2 的刺激表明 ,病人对外界钙激动剂更敏感。胰岛素能刺激胞内 Ca2 浓度升高 ,其作用机制是激活钙离子通道。肾上腺素不仅能激活钙离子通道 ,也能促使钙从钙池中释放。上述研究也表明 Fluo- 3是有效的研究胞内 Ca2 变化的荧光探针。 2型糖尿病的发生可能与 Ca2 浓度升高有关 ,患者对外界钙激动剂更敏感

     

    Abstract: The purpose of the research was to determine the intracellular calcium of patients of NIDDM. We also analyzed the mechanism of several drugs such as Adrenaline, Dobutamine, Bay K 8644 and Insulin which stimulated calcium release of NIDDM, using Fluo 3 AM
